Keap1hasbeenshowntointeractwithNrf2,amasterregulatoroftheantioxidantresponse,whichisimportantfortheameliorationofoxidativestress.Underquiescentconditions,Nrf2isanchoredinthecytoplasmthroughbindingtoKeap1,which,inturn,facilitatestheubiquitinationandsubsequentproteolysisofNrf2.SuchsequestrationandfurtherdegradationofNrf2inthecytoplasmaremechanismsfortherepressiveeffectsofKeap1onNrf2.

BecauseNrf2activationleadstoacoordinatedantioxidantandanti-inflammatoryresponse,andKeap1repressesNrf2activation,Keap1hasbecomeaveryattractivedrugtarget.Syntheticoleanetriterpenoidcompounds,knownasantioxidantinflammationmodulators(AIMs)havebeenshowntoblockKeap1-dependentNrf2ubiquitinationandleADIngtothestABIlizationandnucleartranslocationofNrf2andsubsequentinductionofNrf2targetgenes.
